Simple test of EDC/NHS coupling efficiency

Our new Amine Coupling Test Kit offers a simple and accurate way of determining the coupling efficiency of EDC/NHS amine coupling reactions.

Covalent coupling of amine groups to carboxyl groups is swiftly and conveniently mediated through the use of EDC and NHS coupling reagents. The amine coupling is frequently used for conjugation of biomolecules to e.g. gold and latex nanoparticles, biotin, fluorescent dyes, or Episentec™ dyes for Label-Enhanced SPR. More specifically, the amine coupling is often used for immobilizing biomolecules to sensor surfaces, e.g. in SPR methods like Biacore™.

Using our Amine Coupling Test Kit B9005, the coupling efficiency can now be accurately determined with a simple spectrophotometric mix-and-read method.

  • Quality control of EDC/NHS
    • The actual quality of EDC and NHS may vary, leading to varying coupling efficiencies and sometimes insufficient coupling. It is known that the EDC/NHS quality may vary between different vendors and even between different batches.
  • Stability testing of EDC/NHS
    • EDC and NHS are quite unstable. They are prone to hydrolysis by water and it’s difficult to deem the quality of reagents in opened containers. Once dissolved in water, EDC and NHS may be stored for months in the freezer, but it’s recommended to check the quality before using such reagents.
  • Evaluation and optimization of coupling conditions
    • The reaction conditions used for amine coupling may vary. There are many different protocols published, suggesting e.g. different solvents, pH, and reaction times. Using our Amine Coupling Test Kit, different reaction conditions can easily be evaluated and optimized.
